COZMOS is a Phase I/Ib study evaluating combined 5’azacitidine and carboplatin in children with recurrent or refractory paediatric brain and solid tumours. The study is based on the rationale that many paediatric tumours demonstrate altered epigenetic landscapes and DNA methylation patterns.
The Phase I component evaluates escalating doses of carboplatin in combination with 5’azacitidine to determine the maximum tolerated dose. The Phase Ib expansion cohort includes up to 30 patients with recurrent ependymoma, comprising recurrent posterior fossa and supratentorial ependymoma cohorts.
The study assesses safety, tolerability, dose-limiting toxicities, and preliminary anti-tumour activity of this combination approach.
