The TarGeT-D study tests the effectiveness of olutasidenib and temozolomide in patients with a specific genetic change called an IDH1 mutation in newly diagnosed High Grade Glioma (HGG) or DIPG. This study tracks how the disease responds or resists treatment by looking at laboratory findings, brain scans, and patient feedback. It also investigates “pseudoprogression,” which is when a brain scan shows changes that might look like tumour growth but are actually the brain healing from treatment. By using advanced imaging, the goal is to refine personalised treatment strategies for this specific genetic group of paediatric brain tumours.
TarGeT-D is a molecularly stratified therapeutic arm of the CONNECT TarGeT umbrella protocol. This Phase II study evaluates the efficacy and safety of olutasidenib, a selective oral inhibitor of mutated IDH1, administered in combination with temozolomide for paediatric and young adult patients with newly diagnosed IDH1-mutant HGG or DIPG. The primary objective is to determine if this targeted combination improves clinical outcomes following radiotherapy. Correlative science includes analysing molecular markers of response, validating findings via neuroimaging, and evaluating the incidence of pseudoprogression to refine precision oncology strategies for this genetic subset.
